Pregnenolone, anonymous and unheralded as it may be, is the Mother of all steroid hormones. It is derived from cholesterol. From there it’s converted into testosterone, progesterone, estrogen and cortisol, among other important biochemicals. Even vitamin D, the body’s homemade essential nutrient, is a derivative of the under-appreciated biochemical.

Pregnenolone itself, in its raw unconverted form, can be considered the quintessential biochemical of youth. Kids and teenagers make loads of it. While pregnenolone on its own may not have many hormone properties, it is able to improve the activity of and stabilize the effects of other hormones, including cortisol, testosterone and estrogen. As we get older, our pregnenolone levels tend to decrease. This is one of the reasons for the ever increasing degenerative impact of stress associated with aging.

The stabilizing effect is especially prominent when it comes to neurons, which are among the most active cells in the body. This can make pregnenolone a very helpful supplement for all brain and mental health issues, including anxiety, insomnia and depression. Because the production of pregnenolone drops as we age, supplementation may be something to think about for older folks who are experiencing memory problems, mental health issues and even dementia. It protects brain cells from excitotoxins and fatigue induced injury, as well as having a calming effect that may be associated with its feel-good properties. It has a mellowing effect on nerve cells, while blocking excessive inhibition of the brain’s sleepy-time amino acids, glycine and GABA, helping to balance chemistry in the gray matter.

Pregnenolone is also important for cognitive functioning. Declining levels associated with aging are linked to memory and learning problems. A 2003 study in the journal, “Progress in Neurobiology”, found that pregnenolone reduced the age-related decline in nerve cell growth in rats. Other studies show that aging lab animals given supplemental pregnenolone have improvements in memory and performance. Also, according to the National Institute of Health, pregnenolone deficiencies may be be one of the factors responsible for the cognitive decline related to normal aging or associated with Alzheimer's disease.

One of the more intriguing features of pregnenolone is that its calming effect directly antagonizes some of the side effects associated with anxiety medication, particularly benzodiazepines e.g. Valium and Xanax.

According to a 2004 article published in the journal “Psychneuroendocrinology”, pregnenolone pre-treated subjects showed a significant and clinically relevant reduction in sedation following administration of Valium. It can be considered at least a partial blocker of benzodiazepines. This raises the possibility of it helping people withdraw from addiction to the commonly used anti-anxiety drugs.

Pregnenolone also has an attenuating effect on natural stress hormones. This makes it a potentially important supplement for anyone dealing with the effects of excess cortisol, including skin effects like oily skin and acne. Likewise, there are potential benefits for estrogenic health issues, including autoimmune disease and also for PMS. I have personally seen some remarkable benefits for women suffering from premenstrual anxiety issues and emotional instability with a 100 mg daily dose. The only downside to Pregnenolone supplementation is a little drowsiness if you take too much. Interestingly at just the right dose however, Pregnenolone can have an energizing effect.

There’s an interesting relationship between marijuana and pregnenolone. According to a January 2014 article in Science Magazine, pregnenolone was described as being able to mitigate some of the effects of cannabis toxicity. When lab mice were exposed to tetrahydrocannabinol (THC), the pot plant’s most significant active material, their brains initiated a 3,000 percent increase in the production of pregnenolone. Scientists are looking to exploit this relationship, hoping that pregnenolone may be useful in treating marijuana induced brain trauma and perhaps even benefit patients dealing with addiction.

Pregnenolone’s benefits are not only for the internal health of the body; it also has cosmetic benefits. Recently, a flurry of research has shown that various cells in the skin are important producers of many steroid hormones. This seems logical given the skin dynamism protective role. Skin cells called keratinocytes are known producers of pregnenolone, perhaps to leverage the hormone’s anti-inflammatory and anti-stress benefits. Pregnenolone’s cortisol and testosterone balancing effects may be helpful for slowing down the production of skin oils and the formation of acne blemishes. Anecdotally, some people claim that pregnenolone has tightening effect on the skin, which may be the result of the hormone’s effects on skin muscle.

Pregnenolone is readily available via health food stores and the internet. Which means you can do your own replacement therapy by taking 100 to 300 mg a day. When taken orally, even in the powdered form, it is absorbed fairly well. It has a long-lasting effect; in addition to the dose itself, supplemental pregnenolone may be able to help your body’s produce its own natural version of this important hormonal substance.
